目的研究8-姜酚处理小鼠脾淋巴细胞和脾巨噬细胞等免疫细胞的影响。方法分离并培养小鼠脾淋巴细胞和脾巨噬细胞,用MTT比色法测定8-姜酚对小鼠脾淋巴细胞及经革兰氏阴性菌细胞壁脂多糖(LPS)和刀豆蛋白A(ConA)诱导的脾淋巴细胞增殖能力的影响;用巨噬细胞吞噬中性红试验考察8-姜酚对脾巨噬细胞及经LPS和ConA活化的脾巨噬细胞吞噬功能的影响。结果 8-姜酚(终浓度3.125、6.25、12.5、25、50和100μg/ml)对检测小鼠脾淋巴细胞增殖能力和脾巨噬细胞吞噬功能均有抑制作用,其半数抑制浓度(IC50)分别为3.795和0.185 mg/ml;对LPS和ConA诱导的脾淋巴细胞增殖能力和脾巨噬细胞的吞噬功能有显著的抑制作用(P<0.05)。结论在该试验条件下,8-姜酚对小鼠脾淋巴细胞和巨噬细胞及其经LPS和ConA活化后的增殖与吞噬作用均有抑制作用。
Objective To study the effects of 8-gingerol on immune cells such as splenic lymphocytes and splenic macrophages in mice. Methods Splenic lymphocytes and splenic macrophages were isolated and cultured in vitro. MTT assay was used to determine the effect of 8-gingerol on lipopolysaccharide (LPS) and concanavalin A ConA) -induced spleen lymphocyte proliferation. The effect of 8-gingerol on phagocytosis of splenic macrophages and splenic macrophages activated by LPS and ConA was investigated by macrophage phagocytosis neutral red assay. Results 8-Gingerol (3.125,6.25,12.5,25,50 and 100μg / ml) had inhibitory effect on the proliferation of splenic lymphocytes and phagocytosis of spleen macrophages in mice. The IC 50 values 3.795 and 0.185 mg / ml, respectively. LPS and ConA could significantly inhibit the proliferation of splenic lymphocytes and the phagocytosis of splenic macrophages (P <0.05). Conclusion Under these experimental conditions, 8-gingerol can inhibit the proliferation and phagocytosis of mouse spleen lymphocytes and macrophages and their activation by LPS and ConA.
