目的:比较野生和栽培金铁锁的镇痛效果及其具镇痛活性的有效部位。方法:提取分离获得野生和栽培金铁锁根部不同极性的化合物,运用醋酸扭体法和热板法研究其对小鼠疼痛的抑制效果。结果:无论是野生还是栽培的金铁锁,其浸膏的镇痛效果相似,均含有高活性的镇痛成分。从化学刺激致痛的抑制效果来看,栽培与野生金铁锁活性成分均存在于正丁醇(扭体抑制率分别为99.1%±3.0%和100.0%±0.0%)、乙酸乙酯(97.9%±6.3%和81.9%±11.7%)和石油醚部分(78.8%±17.7%和70.9%±10.3%),但栽培金铁锁乙酸乙酯部分的镇痛效果(97.9%±6.3%)明显优于野生金铁锁(81.9%±11.7%,P<0.05)。从热刺激致痛模型的镇痛作用来看,野生金铁锁的活性部位为正丁醇部分(30 min痛阈提高率为166.5%±71.6%,60min为186.9%±79.0%),栽培金铁锁的活性部位为正丁醇部分(30 min痛阈提高率为128.3%±53.1%,60 min为136.5±65.2%)和乙酸乙酯部分(30 min为53.7%±24.4%,60 min为186.9%±79.0%)。结论:虽然人工栽培金铁锁的镇痛活性成分与野生药材有所不同,但其镇痛功效与野生药材无明显区别,可作为镇痛药物等效替代野生药材。
OBJECTIVE: To compare the analgesic effect of wild and cultivated gold iron lock and its effective site with analgesic activity. Methods: The compounds with different polarities of wild and cultivated luteolin were extracted and isolated. The inhibitory effect on the pain of mice was studied by acetic acid writhing method and hot plate method. Results: Both wild and cultivated Jin Tie Suo, its extract analgesic effect is similar, both contain highly active analgesic ingredients. In terms of the inhibitory effect of chemical stimuli induced pain, the active components of both cultivated and wild Metallopods were found in n-butanol (writhing inhibition rates of 99.1% ± 3.0% and 100.0% ± 0.0%, respectively), ethyl acetate (97.9% ± 6.3%, and 81.9% ± 11.7%) and petroleum ether fractions (78.8% ± 17.7% and 70.9% ± 10.3%), respectively. However, the analgesic effect (97.9% ± 6.3%) of the ethyl acetate- (81.9% ± 11.7%, P <0.05). From the analgesic effect of heat-induced pain model, the active sites of wild gold-iron lock were n-butanol fraction (166.5% ± 71.6% at 30 min and 186.9% ± 79.0% at 60 min) (N = 30) were n-butanol fraction (128.3% ± 53.1% at 30 min and 136.5 ± 65.2% at 60 min) and ethyl acetate fraction (53.7% ± 24.4% at 30 min and 186.9% ± 79.0 at 60 min %). Conclusion: Although the analgesic active ingredients of Artificial Cultivation are different from those of wild medicinal herbs, their analgesic efficacy is not significantly different from that of wild medicinal herbs and can be used as an equivalent substitute of wild medicinal herbs for analgesics.
