目的探索外周血自然杀伤T(NKT)细胞比例变化与非酒精性脂肪性肝病(NAFLD)的关系。方法应用流式细胞术检测60例NAFLD患者和60名年龄、性别配对的正常对照者外周血NKT细胞比例,同时比较两组间各项临床变量的差异并分析各项临床变量与外周血NKT细胞比例变化的关系.Logistic回归分析筛选NAFLD发病的危险因素。结果正常对照组外周血NKT细胞比例的中位数及四分位数区间为1.48%(1.23%~1.98%),NAFLD组为1.12%(0.87%~1.54%),两组差异有统计学意义(P<0.001)。Spearman相关分析发现外周血NKT细胞比例与体重指数、腹围、丙氨酸转氨酶水平呈负相关(r值分别为-0.322、-0.237、-0.217:P值分别为0.001、0.021、0.035)。Logistic回归分析发现外周血NKT细胞比例、体重指数、天冬氨酸转氨酶、空腹血糖与NAFLD的发病危险因素有关(OR值分别为0.107、2.991、1.148和3.133.P值分别为0.023、0.003、0.026和0.013)。结论NAFLD患者外周血NKT细胞比例是下降的,且该比例下降可能是NAFLD发病的危险因素之一。
Objective To explore the relationship between the proportion of natural killer T (NKT) cells in peripheral blood and non-alcoholic fatty liver disease (NAFLD). Methods Flow cytometry was used to detect the percentage of NKT cells in peripheral blood of 60 NAFLD patients and 60 normal controls matched for age and sex. The differences of clinical variables between the two groups were compared and analyzed with the correlation between clinical variables and peripheral blood NKT cells Logistic regression analysis was used to screen the risk factors of NAFLD. Results The median and interquartile range of NKT cells in the control group were 1.48% (1.23% -1.98%) and 1.12% (0.87% -1.54%) in the NAFLD group, with significant difference between the two groups (P <0.001). Spearman correlation analysis showed that the proportion of peripheral blood NKT cells and body mass index, abdominal circumference, alanine aminotransferase levels were negatively correlated (r values were -0.322, -0.237, -0.217: P values were 0.001,0.021,0.035, respectively). Logistic regression analysis showed that the proportion of NKT cells in peripheral blood, body mass index, aspartate aminotransferase, fasting blood glucose and NAFLD risk factors (OR = 0.107,2.991,1.148 and 3.133.P values were 0.023,0.003,0.026 And 0.013). Conclusion The proportion of NKT cells in peripheral blood of NAFLD patients is decreased, and the decrease may be one of the risk factors of NAFLD.
