【目的】研究紫心马铃薯品种黑美人的组织培养技术,为其种薯工厂化生产提供技术支持。【方法】以紫色马铃薯品种黑美人的顶芽及不同部位腋芽为外植体,探讨0.1%HgCl2对外植体灭菌、6-BA对初代培养、6-BA和PP333组合对增殖培养、NAA对生根、糖对结薯等过程的影响。【结果】在4.0~9.0min范围内,以0.1%升汞溶液对顶芽、第2段腋芽、第3段腋芽及以下芽分别处理5、6、8 min的灭菌效果最好。在MS培养基中添加0~3.0 mg/L 6-BA,顶芽、第2段腋芽、第3段腋芽及以下芽的适宜初代培养基分别为MS+1.0 mg/L 6-BA、MS+1.5 mg/L 6-BA、MS+2.0 mg/L6-BA。在MS培养基中添加0~3.0mg/L 6-BA和0~0.4 mg/L PP333,较适宜的增殖培养基为MS+3.0 mg/L 6-BA+0.2 mg/L PP333,最高增殖倍数为7.0。在0~0.6mg/L NAA内,较适宜的生根培养基为MS+0.4 mg/L NAA,最高生根率达100.0%。MS培养基中添加30.0~80.0 mg/L糖,较适宜的结薯培养基为MS+60.0 g/L糖,最高结薯率达93.3%。【结论】成功构建了紫色马铃薯黑美人的再生繁殖体系,获得无菌苗及试管薯,该繁殖技术可用于其种薯工厂化生产。
【Objective】 The purpose of this study was to study tissue culture techniques of purple sweet potato variety Black Beauty and to provide technical support for its seed potato production. 【Method】 The apical buds and axillary buds of different parts of purple potato were used as explants to investigate the effects of 0.1% HgCl2 on explants, 6-BA on primary culture, 6-BA and PP333 on proliferation, NAA Roots, sugar on the tuber and other processes. 【Result】 The results showed that the sterilization effect of 0.1% mercuric chloride solution on the top buds, the second axillary buds, the third axillary buds and the following buds for 5, 6, 8 min were the best in the range of 4.0-9.0 min. The suitable primary culture medium supplemented with 0-3.0 mg / L 6-BA, the top bud, the second axillary bud, the third axillary bud and the following buds were MS + 1.0 mg / L 6-BA and MS + 1.5 mg / L 6-BA, MS + 2.0 mg / L 6-BA. The best medium for proliferation was MS + 3.0 mg / L 6-BA + 0.2 mg / L PP333 with MS medium supplemented with 0-3.0 mg / L 6-BA and 0-0.4 mg / L PP333. 7.0. In 0 ~ 0.6mg / L NAA, the most suitable rooting medium was MS + 0.4 mg / L NAA with the highest rooting rate of 100.0%. MS medium supplemented with 30.0 ~ 80.0 mg / L sugar, the optimum medium for tuber culture was MS + 60.0 g / L sugar, the highest tuber yield reached 93.3%. 【Conclusion】 The regenerated and reproductive system of purple potato black beauty was successfully constructed, and sterile germ and tube-tuber were obtained. The breeding technology can be used for the seed production of its seed potato.
