目的构建反义胰岛素样生长因子-1(IGF-1)表达质粒(pCEP4-IGF-1A)治疗大鼠C6脑胶质瘤。方法RT-PCR扩增300bpIGF-1cDNA,测序,构建反义IGF-1表达质粒,体外转染C6细胞,观察其体外增殖速率及对C6胶质瘤的治疗作用。结果反义IGF-1序列正确,反义IGF-1A转染的C6细胞生长明显受抑,体内成瘤性消失,荷瘤鼠肿瘤全部消失,6个月观察期内肿瘤无复发。结论反义IGF-1表达质粒能抑制C6细胞体外生长,体内杀伤肿瘤效果明显。
Objective To construct antisense insulin-like growth factor-1 (IGF-1) expression plasmid (pCEP4-IGF-1A) for rat C6 glioma. Methods The cDNA of 300bp IGF-1 was amplified by RT-PCR and sequenced. Antisense IGF-1 expression plasmid was constructed and transfected into C6 cells in vitro. The proliferation rate and the therapeutic effect on C6 glioma were observed. Results The antisense IGF-1 sequence was correct. The growth of C6 cells transfected with antisense IGF-1A was significantly inhibited and the tumorigenicity disappeared in vivo. All the tumors in tumor-bearing mice disappeared, and the tumor did not recur in the 6-month observation period. Conclusion Antisense IGF-1 expression plasmid can inhibit the growth of C6 cells in vitro and kill tumor in vivo.
