为了观察蛋白酪氨酸激酶(PTK) 是否参与肥大细胞活化的信号转导, 应用流式细胞仪和免疫细胞化学方法, 观察了致敏肥大细胞RBL2H3 在抗原二硝基苯酚牛血清白蛋白(DNPBSA) 激发下, 胞浆中磷酸酪氨酸浓度的改变。结果发现, 致敏RBL2H3 细胞在DNPBSA激发下, 迅速活化, 由异硫氰酸酯荧光素标记的二抗绿荧光强度为4-46 ±0-06, 明显高于对照组1-84±0-02, ( P< 0-01 ), 绿荧光主要分布于胞浆中, 说明PTK 参与肥大细胞活化的信号转导
In order to observe whether protein tyrosine kinase (PTK) is involved in the signal transduction of mast cell activation, flow cytometry and immunocytochemistry were used to observe the sensitized mast cells RBL 2H3 in the antigen dinitrophenol bovine serum Protein (DNPBSA) excitation, changes in the concentration of phosphotyrosine in the cytoplasm. The results showed that sensitized RBL 2H3 cells activated rapidly in DNPBSA excited by fluorescein isothiocyanate-labeled second antibody green fluorescence intensity of 4-46 ± 0-06, was significantly higher than the control group 1- 84 ± 0-02, (P <0-01). The green fluorescence mainly distributed in the cytoplasm, indicating that PTK is involved in the signal transduction of mast cell activation
