目的 研究羟自由基 (一类机制 )、单线态氧 (二类机制 )在DMMB与M0 0 7光化学灭活病毒和对红细胞损伤中的作用。方法 在光化学反应体系中加入羟自由基猝灭剂甘露醇、甘油 ,单线态氧猝灭剂色氨酸和叠氮钠 ,试验它们对灭活Phi6噬菌体的抑制能力以及对红细胞溶血、钾泄漏的防护作用。结果 单线态氧猝灭剂可以明显抑制DMMB和M0 0 7光化学灭活Phi6噬菌体的能力 ,羟自由基猝灭剂对灭活Phi6噬菌体影响很小 ,而两类猝灭剂都对红细胞溶血有一定的防护作用。结论 DMMB、M0 0 7主要通过二类机制来灭活病毒 ,一、二类机制都参与了对红细胞的损伤。
Objective To investigate the effects of hydroxyl radical (a kind of mechanism), singlet oxygen (a second kind of mechanism) on the photochemical inactivation of virus by DMMB and M0 0 7 and on erythrocyte injury. Methods In the photochemical reaction system, mannitol, glycerol, singlet oxygen quencher tryptophan and sodium azide were added as quenching agents, and their inhibitory activity on inactivated Phi6 phage and their effects on erythrocyte hemolysis and potassium leakage Protective effect. Results The singlet oxygen quencher significantly inhibited the photochemical inactivation of Phi6 phage in DMMB and M0 0 7. The hydroxyl radical quencher had little effect on inactivated Phi6 phage, but both of the two quenchers had a definite effect on erythrocyte hemolysis The protective effect. Conclusion DMMB and M0 07 mainly inactivated virus by two kinds of mechanisms. One or two kinds of mechanisms involved in the damage of erythrocytes.
