以自提鳕鱼皮明胶以及海藻酸钠为原料, 经戊二醛交联制备了复合止血敷料, 并对其物理性能、微观结构和肝脏相容性进行了研究.结果表明, 在体外凝血时间中, 鳕鱼皮明胶的最佳止血浓度为15mg/mL, 止血时间为 (151±12) s, 说明鳕鱼皮明胶具有良好的止血性能.综合抗张强度、断裂伸长率、吸水倍数、保湿率和交联度5项物理指标, 确定了复合止血敷料中明胶与海藻酸钠的最优复配比为15∶8.扫描电镜图表明海藻酸钠的添加改善了止血敷料的微观结构, 使其具有孔径大小 (100~200μm) 一致且分布均匀的多孔结构.在肝脏相容性实验中, 复合止血敷料在植入后第8周时完全降解, 没有引起严重的炎症反应, 表明鳕鱼皮明胶复合止血敷料具有良好的体内降解性, 是1种潜在的可吸收医用材料.“,”The aim of this study was to prepare a composite hemostatic sponge based on gelatin from Pacific cod (Gadus macrocephalus) skin and sodium alginate with glutaraldehyde as crosslinking agent, and its physical properties, microstructure and liver compatibility were studied.The results showed that the best hemostatic concentration of gelatin from Pacific cod skin was 15 mg/mL in the conagulation time in vitro, and the hemostatic time was (151±12) s.Based on physical indexes including tensile strength, elongation at break, water absorption, water retention and crosslinking degree, the best ratio between gelatin and sodium alginate was 15∶8.Scanningelectron microscope (SEM) showed that the composite hemostatic sponge had a porous structure with uniform pore size and distribution.In the experiment on liver compatibility, the composite hemostatic sponge was completely absorbed after eight weeks of implantation, and it didn't cause severe inflammatory response, indicating that it had good biodegradability in vivo.Thus, the composite hemostatic sponge based on gelatin from Pacific cod skin would be a potential candidate for wound dressing.
