解毒化瘀颗粒对急性肝衰竭大鼠细胞因子及肝组织的影响

目的:从炎症因子角度探讨解毒化瘀颗粒对急性肝衰竭大鼠免疫网络的细胞因子调节作用和对肝组织的影响。方法:取Wistar大鼠120只,取正常组20只,其余大鼠采用D-氨基半乳糖(D-Gal N)+脂多糖(LPS)联合制备大鼠急性肝衰竭模型,造模成功的大鼠分别分为模型组,解毒化瘀颗粒低、中、高剂量组(9.1,28.76,57.55 g·kg~(-1)),E5531组(内毒素拮抗剂,10 mg·kg~(-1)),每组20只,造模前5 d开始ig给药,正常组与模型组均给予等量蒸馏水,2次/天,造模后给药48 h后处死大鼠,酶联免疫吸附测定(ELISA)法检测各组大鼠血清肿瘤坏死因子-α(TNF-α),白细胞介素-6(IL-6),IL~(-1)0水平;采用苏木素-伊红(HE)染色观察肝脏病理组织学改变。结果:与正常组比较,模型组大鼠血清TNF-α,IL-6水平明显升高,IL~(-1)0水平明显降低(P<0.05),模型组大鼠肝组织明显的炎性细胞浸润,病理损伤较为明显;与模型组比较,解毒化瘀颗粒低、中、高剂量组TNF-α,IL-6水平及显著降低,IL~(-1)0水平表达升高(P<0.05),肝组织病理结果提示解毒化瘀颗粒低、中、高剂量组的肝组织损伤程度均较模型组减轻,有显著性差异(P<0.05)。结论:解毒化瘀颗粒通过减少促炎因子的水平、抑制促炎因子表达,提高抗炎因子的水平及表达来改善患者全身炎症反应、保护肝细胞功能,解毒化瘀颗粒拮抗急性肝衰竭大鼠的作用机制之一。 OBJECTIVE: To explore the regulatory effect of Jiedu Huayu granules on the immune cytokines in acute liver failure rats and its effect on liver tissue from the perspective of inflammatory factors. Methods: A total of 120 Wistar rats were selected and 20 rats in normal group were used. The rats in the other groups were treated with D-Gal N and LPS for acute hepatic failure model. The rats were divided into model group, low, middle and high dosage of Jiedu Huayu granules (9.1,28.76,57.55 g · kg -1), E5531 group (10 mg · kg -1 ), With 20 rats in each group. The animals were dosed 5 days before the model was established. Rats in the normal group and model group were given equal volume of distilled water twice a day for 48 hours after the model was established. Enzyme-linked immunosorbent assay Serum levels of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) and IL-1 0 were detected by ELISA. Histopathological changes of liver were observed by staining. Results: Compared with the normal group, the levels of TNF-α and IL-6 in the model group were significantly increased, and the level of IL-1 (0) was significantly decreased (P <0.05) Compared with the model group, the levels of TNF-α, IL-6 and the level of IL-1 in the low, medium and high dosage groups of Jiedu Huayu granules were significantly increased (P < 0.05). The pathological results of liver tissue suggested that the degree of liver damage of low, middle and high dosage group of Jiedu Huayu Granules was lighter than that of model group, with significant difference (P <0.05). Conclusion: Jiedu Huayu Granule can prevent liver injury by reducing the levels of proinflammatory cytokines, inhibiting the expression of proinflammatory cytokines, increasing the level and expression of anti-inflammatory cytokines in patients with systemic inflammatory response and protecting liver cells. One of the mechanisms of action.
