目的研究糖基化终产物(AGEP)对人脐静脉内皮细胞二酰基甘油(Dia)水平的影响和维生素E、氨基胍对AGEP升高内皮细胞Dia水平的干预作用。方法采用放射酶标记、簿层层析、放射自显影技术测定细胞中Dia含量。结果AGEP-BSA孵育的内皮细胞Dia水平出现双时相性变化,并随AGEP-BSA浓度、糖基化程度和修饰AGEP-BSA的糖浓度增加,Dia水平亦明显增高、维生素E和氨基胍可明显减弱AGEP-BSA升高Dia水平的效应。结论AGEP-BSA能升高人脐静脉内皮细胞中Dia含量,维生素E和氨基肌明显降低AGEP升高的细胞内Dia水平。
Objective To investigate the effect of advanced glycation end products (AGEP) on the level of diacylglycerol (Dia) in human umbilical vein endothelial cells (ECs) and the effect of vitamin E and aminoguanidine on the level of DiaE in endothelial cells stimulated by AGEP. Methods The content of Dia was determined by radioimmunoassay, scanning electron microscopy and autoradiography. Results The Dia level of endothelial cells in AGEP-BSA incubated with different concentrations of AGEP-BSA, the degree of glycosylation and the concentration of AGEP-BSA increased the level of Dia, and the levels of Dia were also significantly increased. Vitamin E and aminoguanidine were significantly Attenuate the effect of AGEP-BSA on increasing Dia level. Conclusion AGEP-BSA can increase the content of DiaE in human umbilical vein endothelial cells. Vitamin E and aminomycin significantly reduce the intracellular Dia level of AGEP.
