金钗石斛多糖对大鼠混合培养皮层细胞炎症相关因子表达的抑制作用

目的:探索金钗石斛多糖(NDP)对脂多糖(LPS)作用的新生大鼠大脑皮层胶质细胞-神经元混合培养体系的保护作用。方法:制备新生大鼠大脑皮层胶质细胞-神经元混合培养体系,NDP预处理后LPS刺激,观察细胞生长情况并采用Western blot法检测体系中炎症相关因子IL~(-1)β,TNF-α,COX-2和IL-6的蛋白表达及相关调控信号p-NF-κB,p-p38,p-JNK和p-ERK的水平。结果:NDP作用于LPS刺激的大鼠皮层胶质细胞-神经元混合培养体系后,较单用LPS的模型组相比炎症相关因子IL~(-1)β,TNF-α,COX-2和IL-6的蛋白表达及相关调控信号p-NF-κB,p-p38,p-JNK和p-ERK的水平明显降低。结论:NDP可抑制LPS诱导的新生大鼠皮层胶质细胞-神经元混合培养体系炎症相关因子的产生。 Objective: To explore the protective effect of Dendrobium nobile polysaccharide (NDP) on glial cell neuron culture in neonatal rats with lipopolysaccharide (LPS). Methods: Cultured neonatal rat cortical glial cells - neurons mixed culture system, NPS pretreatment LPS stimulation to observe the cell growth and Western blot detection of inflammatory cytokines in the system of IL - (-1) beta, TNF- α, COX-2 and IL-6 protein levels and the related regulatory signals of p-NF-κB, p-p38, p-JNK and p-ERK. Results: Compared with the LPS alone group, NDP could significantly decrease the levels of IL-1β, TNF-α, COX-2 and TNF-α in LPS-stimulated rat cortical neurons mixed culture system IL-6 protein expression and related regulatory signals p-NF-κB, p-p38, p-JNK and p-ERK levels were significantly lower. Conclusion: NDP can inhibit the LPS-induced neonatal rat cortical neurons mixed culture system inflammation-related factors.
