基于UPLC/QTOF-MS技术的补骨脂水提物对大鼠血浆代谢组学研究

目的:研究补骨脂水提物对大鼠血清内源性代谢产物的影响,并从中发现相关代谢标志物。方法:雄性SD大鼠每天1次分别给予补骨脂水提物0.54,1.08和1.62 g/kg,连续给药2周,末次给药后,禁食24h,采集血液,应用超高效液相色谱/四级杆飞行时间质谱(UPLC-Q-TOF-MS)分析样品,Mass Lynx采集并处理数据,采用偏最小二乘判别分析(PLS-DA)方法,分析各组大鼠血清内源性代谢产物的差别,通过正交偏最小二乘法(OPLS-DA)、变量重要程度(VIP)筛选潜在生物标志物并测定其相对含量。结果:与空白组相比,各给药组吲哚、L-苯丙氨酸和植物鞘氨醇含量呈明显上升,乳酸和芽子碱甲酯含量呈明显下降(P<0.05,P<0.01)。结论:补骨脂水提物给药2周后,大鼠血清内源性物质发生显著改变,补骨脂水提物发挥药效或毒效的机制可能是通过调节氨基酸代谢、磷脂代谢以及糖酵解过程来实现的。 Objective: To study the effect of aqueous extract of Psoralen on serum endogenous metabolites in rats and to find related metabolic markers. Methods: Male SD rats were given 0.54, 1.08 and 1.62 g / kg water extract once a day for 2 weeks continuously. After the last administration, the rats were fasted for 24 hours. CONCLUSION: After 2 weeks of administration of Psoraleal water extract, the endogenous substances in rat serum change significantly. The mechanism of efficacy or toxic effect of aqueous extract of Psoralen may be through the regulation of amino acid metabolism, phospholipid metabolism and sugar Glycolysis process to achieve.
