目的确定超声波法提取平菇多糖的最佳工艺。方法采用超声波法提取平菇多糖,利用苯酚硫酸法显色,在490 nm处测定吸光度值,并以正交试验设计优化工艺条件。结果平菇多糖的最佳提取工艺条件为料液比1:30、超声时间60 min、提取温度40℃、超声功率为175 W。结论本实验平菇多糖多糖提取方法及含量测定操作简便,结果准确,重现性好,可用于平菇多糖的提取与含量测定。
Objective To determine the optimum extraction process of polysaccharides from Pleurotus ostreatus by ultrasonic method. Methods The polysaccharides of Pleurotus ostreatus were extracted by ultrasonic method, and the absorbance value was measured at 490 nm by phenol sulfuric acid method. The orthogonal design was used to optimize the process conditions. Results The optimal extraction conditions of polysaccharides from Pleurotus ostreatus were as follows: the ratio of material to liquid was 1:30, the ultrasonic time was 60 min, the extraction temperature was 40 ℃ and the ultrasonic power was 175W. Conclusion Pleurotus ostreatus polysaccharide polysaccharide extraction method and determination of the simple, accurate, reproducible, and can be used for mushroom polysaccharide extraction and determination.
