探索黑翅土白蚁表皮碳氢化合物的固相微萃取条件。应用固相微萃取、气相色谱和气相色谱-质谱联用等技术,从样品量、萃取温度、萃取时间、萃取头选择、载气选择和萃取方式选择6个因素,应用峰总面积、峰个数及保留时间为指标,评价固相微萃取条件优化的效果。80头工蚁放入15 m L样品瓶,用100μm PDMS萃取头在60℃下顶空萃取90 min为黑翅土白蚁表皮碳氢化合物检测的最佳萃取条件。相同实验条件下,分别用氮气或氦气作为载气进行表皮碳氢化合物分析,峰形和出峰顺序上无差异,但氮气作为载气时出峰时间稍延后。获得的固相微萃取工作条件优化不仅可用于黑翅土白蚁表皮碳氢化合物的萃取分析,也可作为相关研究参考。氮气或氦气作为载气对实验结果无明显影响。萃取效果与单位容积样品量之间不呈简单的线性关系,其机理有待进一步研究。 Exploration of solid phase microextraction conditions for the ternary epidermis of the black-winged termites. Six factors (sample size, extraction temperature, extraction time, selection of extraction heads, carrier gas selection and extraction methods) were selected by using solid phase microextraction, gas chromatography and gas chromatography-mass spectrometry. Number and retention time as an indicator to evaluate the effect of optimization of solid-phase microextraction conditions. 80 workers were placed in a 15 mL sample vial and extracted with 100 μm PDMS headspace at 60 ℃ for 90 min to obtain the best extraction conditions for the detection of epidermal hydrocarbons in the black-winged termites. Under the same experimental conditions, epidermal hydrocarbon analysis was conducted using nitrogen or helium as the carrier gas respectively. There was no difference in peak shape and peak order, but the peak time was slightly delayed when nitrogen was used as the carrier gas. The obtained solid phase microextraction working conditions optimization can not only be used for extraction and analysis of ternary epidermis hydrocarbons of the black-winged termites, but also can be used as reference for the relevant research. Nitrogen or helium as the carrier gas has no significant effect on the experimental results. There is no simple linear relationship between extraction efficiency and sample volume per unit volume, and its mechanism needs further study.
