目的 :通过提高肿瘤细胞活性氧 (ROS)水平 ,探讨增加三氧化二砷 (As2 O3)促进细胞凋亡的敏感性。方法 :As2 O3与天然醌类物质大黄素和醌类氧化酶抑制剂双香豆素合用 ,作用于 He L a细胞 ,分别以 MTT法和流式细胞仪检测细胞活力、细胞凋亡率和活性氧水平。结果 :大黄素 10μm ol/ L通过提高 ROS增强了 He L a细胞对 As2 O3的敏感性 ,双香豆素进一步增强了这一作用。结论 :提高肿瘤细胞 ROS水平可能是提高 As2 O3疗效的一种手段。
OBJECTIVE: To explore the sensitivity of increasing the arsenic trioxide (As 2 O 3) to apoptosis by increasing the level of reactive oxygen species (ROS) in tumor cells. Methods: As2 O3 was used in combination with natural quinone emodin and quinone oxidase inhibitor diclofenac, and the effects of As2O3 on HeLa cells were detected by MTT assay and flow cytometry respectively. Cell viability, apoptosis rate and activity Oxygen levels. RESULTS: Emodin 10 μmol / L increased the sensitivity of HeLa cells to As2 O3 by increasing ROS, and dicoumarol further enhanced this effect. Conclusion: Increasing the level of ROS in tumor cells may be a means to improve the efficacy of As 2 O 3.
