本文应用同位素双标记示踪方法、巨噬细胞功能测定和斑点杂交力法,观察了大槐合剂对实验性高胆固醇血症大鼠LDL清除的受体途径和非受体途径(单核巨噬细胞系统)的影响。与造型组比较,大槐合剂组LDL经受体途径和非受体途径的部分清除率(FCR)均显著增加(P<0.01,P<0.05),非受体途径的第二指数曲线半衰期(t1/2)明显缩短(P<0.05)。单核巨噬细胞系统吞噬功能显著增强,吞噬指数(K,K’)和校正吞噬指数(α)显著增加(P<0.001,P<0.001,P<0.05),半量清除时间(t1/2)显著缩短(P<0.001)。肝胞液LDL受体mRNA表达的相对水平显著增加(P<0.05)。结果提示:大槐合剂可同时促进受体和非受体途径清除LDL的作用。为通过刺激单核巨噬细胞系统功能以治疗纯合子家族性高胆固醇血症(FH)的可能性提供依据。
