目的:建立一种基于蛋白芯片技术的抗体分析新方法。方法:将待分析的抗体制备成抗体芯片,并与生物素标记的抗原反应,然后经银增强法显色,获得可直接观察的微阵列芯片可视化反应结果。结果:实验中分析了10种18株抗体的活性和特异性的强弱,其中6株抗体具有较强的特异性。其中,ⅢC013、HPSⅡB007、HPSⅢB007分别对球蛋白和白蛋白的最低检测浓度可达0.4μg/L。结论:该方法具有通量化前景,且具有微量化、操作简便快捷和结果可视化等特点。
Objective: To establish a new method of antibody analysis based on protein chip technology. Methods: The antibody to be analyzed was prepared as an antibody chip, reacted with biotin-labeled antigen, and then color-developed by silver-enhanced method to obtain a directly visualized microarray chip visualization reaction result. Results: In the experiment, the activity and specificity of 10 kinds of 18 kinds of antibodies were analyzed. Among them, 6 kinds of antibodies had strong specificity. Among them, Ⅲ C013, HPS Ⅱ B007, HPS Ⅲ B007 respectively, the lowest detection concentration of globulin and albumin up to 0.4 μg / L. Conclusion: This method has the prospect of flux, and has the characteristics of micro-quantification, simple and quick operation and visualization of results.
