目的掌握复配式粗杂粮对高脂饮食诱导的大鼠胰岛素抵抗的影响。方法40只SD大鼠随机分为阴性对照组(n=10)和高脂造模组(n=30),分别给予基础饲料和高脂饲料,6周后再将高脂造模组分为高脂模型对照组、米面组、粗杂粮组(n=10),提供相应饲料。继续喂养9周后,测定大鼠血糖和胰岛素水平,RT-PCR法测定脂肪组织中过氧化物酶体增殖物激活受体-γ(PPAR-γ)mRNA表达。结果6周高脂饮食后,高脂造模组胰岛素抵抗指数(HOMA-IR)明显升高,造模成功。粗杂粮组大鼠的体重和HOMA-IR显著低于高脂模型对照组和米面组,粗杂粮组PPAR-γmRNA的表达与其他3组比较明显增加(P<0.05)。结论复配式粗杂粮能显著降低高脂饮食引起的胰岛素抵抗大鼠的血糖、胰岛素水平,改善胰岛素敏感性,其机制可能与增加PPAR-γ的表达有关。
Objective To investigate the effects of compound coarse grains on insulin resistance induced by high fat diet in rats. Methods Forty SD rats were randomly divided into negative control group (n = 10) and high fat model group (n = 30). The rats were given basal diet and high fat diet respectively. After 6 weeks, High-fat model control group, rice noodles group, coarse grains group (n = 10), provide the corresponding feed. After 9 weeks of continuous feeding, blood glucose and insulin levels were measured in rats. The expression of peroxisome proliferator activated receptor-γ (PPAR-γ) mRNA in adipose tissue was determined by RT-PCR. Results After 6 weeks of high-fat diet, the insulin resistance index (HOMA-IR) of the high-fat diet group was significantly increased and the model was established successfully. The body weight and HOMA-IR of the coarse grains group were significantly lower than those of the high fat model control group and the rice flour group. The expression of PPAR-γmRNA in the coarse grains group was significantly increased compared with the other three groups (P <0.05). Conclusions Compound coarse grains can significantly decrease the blood glucose and insulin level and improve the insulin sensitivity in insulin resistant rats induced by high fat diet. The mechanism may be related to increasing the expression of PPAR-γ.
