目的探讨青岛市汉坦病毒主要流行株的基因型。方法在青岛5市采集鼠肺标本,应用逆转录聚合酶链反应(RT-PCR)扩增技术及核苷酸序列测定方法对免疫荧光抗原阳性的HV M部分片段扩增并测序,与山东省其他地区代表株及其他部分标准株进行系统发生树及同源性分析。结果分离的5株HV有3株为SEO型,2株为HTN型。从SEO型病毒间的异同来看,青岛市汉坦病毒分离株和临近地区汉坦病毒分离株相近。结论青岛市HV是SEO和HTN混合型疫区;SEO型基因有较高的稳定性。
Objective To investigate the genotypes of major Hantavirus strains in Qingdao. Methods Murine lung samples were collected from 5 cities of Qingdao. The part of immunofluorescence antigen positive HV M fragments was amplified and sequenced by reverse transcription polymerase chain reaction (RT-PCR) and nucleotide sequencing. Compared with Shandong province Phylogenetic trees and homology analysis of representative strains from other regions and other standard strains. Results The results showed that three of the five strains isolated were SEO type and two strains were HTN type. From the similarities and differences between SEO-type viruses, Hantaan virus isolates in Qingdao and similar areas near the Hantavirus isolates. Conclusions Qingdao HV is a mixture of SEO and HTN-infected areas; SEO-type genes have high stability.
