介绍在同一切片上显示两种抗原物质在同一结构内比较分布的一种双重免疫染色技术。实验用过氧化物酶-抗过氧化物酶(PAP)法作为第一染色序列,用碱性磷酸酶标A蛋白(PAAP)法作为第二染色序列,最终分别产生深棕褐色和紫蓝色反应产物。其特点在于利用A蛋白能与IgG抗体分子Fc段牢固结合的分子生物学特性,在进行第二序列反应显色的同时,使第一序列反应产物的染色强度得以增加。结果表明,此技术能反差鲜明地显示含有不同物质的两种细胞在同一区域内的比较分布。
A dual immunostaining technique that shows the distribution of two antigenic species within the same structure on the same slice is introduced. The peroxidase-antiperoxidase (PAP) method was used as the first staining sequence and the alkaline phosphatase A protein (PAAP) method was used as the second staining sequence to produce dark brown and purple-blue reaction product. The utility model is characterized in that by utilizing the molecular biological characteristics that protein A can firmly bind to the Fc fragment of the IgG antibody molecule, the staining intensity of the reaction product of the first sequence is increased while the color of the second sequence reaction is developed. The results show that this technique can clearly show the comparative distribution of two kinds of cells containing different substances in the same area in contrast.
