目的:对匙羹藤花中的有效成分牛弥菜醇A的含量测定进行研究,为科学评价其质量提供科学依据。方法:采用正交实验优选匙羹藤花样品溶液的制备方法,同时采用高效液相色谱法(HPLC)测定牛弥菜醇A的含量,以氨基柱(4.6 mm×250 mm,5μm)为色谱柱,乙腈-水(90∶10)为流动相,流速1.0 mL.min-1,检测波长207 nm,柱温:30℃。结果:匙羹藤花样品溶液的最佳制备方法为150倍50%甲醇超声提取50 min,牛弥菜醇A在0.400 4~20.02μg与峰面积成良好的线性关系(r=0.999 7),平均回收率为98.98%,RSD 1.15%(n=6)。结论:采用HPLC建立的方法准确、快速、灵敏度高、重复性好,可作为匙羹藤花的质量控制方法。
OBJECTIVE: To study the determination of the content of bovine alcohol A in Gymnema sylvestre, and to provide a scientific basis for the scientific evaluation of its quality. Methods: The orthogonal test was used to optimize the preparation of the sample solution of Gymnema sibirica, and the content of bovine serum albumin A was determined by high performance liquid chromatography (HPLC). Amino columns (4.6 mm × 250 mm, 5 μm) , Acetonitrile-water (90:10) as mobile phase, flow rate 1.0 mL · min-1, detection wavelength at 207 nm, column temperature: 30 ℃. Results: The best preparation method for the sample solution of Gymnema sylvestre was 150 times 50% methanol ultrasonic extraction for 50 min. The linear relationship was obtained between 0.400 4 ~ 20.02 μg and peak area of bovine torostatin A (r = 0.999 7) The recovery was 98.98% with a RSD of 1.15% (n = 6). Conclusion: The method established by HPLC is accurate, rapid, sensitive and reproducible. It can be used as quality control method for Gymnema sylvestre.
