目的:观察经微量注射泵给予慢性心衰大鼠侧脑室芪苈强心胶囊对其心功能、心肌组织形态、神经内分泌因子及下丘脑的影响。方法:采用结扎大鼠冠状动脉左前降支建立心衰模型,造模4周后通过侧脑室插管连接微量注射泵给予芪苈强心胶囊溶液(分为0.01mg/kg、0.02mg/kg剂量组),阳性对照组给予CRH竞争性抑制剂(αh-CRH),给药4周后,应用颈动脉插管检测各组大鼠心功能指标,HE染色观察心肌组织形态,酶联免疫吸附试验检测大鼠血清N-端脑钠肽前体(NT-pro BNP)与血浆促肾上腺皮质激素(ACTH)、前列腺素E2(PGE2)、去甲肾上腺素(NE)含量,RT-PCR方法检测下丘脑促肾上腺皮质激素释放激素(CRH)mRNA与肿瘤坏死因子-α(TNF-α)mRNA的表达。结果:芪苈强心胶囊溶液0.01mg/kg组、芪苈强心胶囊溶液0.02mg/kg组与CRH竞争性抑制剂组均可改善慢性心衰大鼠心功能,减轻心肌组织形态的改变,降低大鼠血清NT-pro BNP与血浆ACTH、PEG2、NE含量,降低下丘脑CRH mRNA与TNF-αmRNA的表达。结论:经侧脑室插管连接微量注射泵给予芪苈强心胶囊可有效改善心衰大鼠心功能和心肌组织形态的改变,降低神经内分泌因子的含量,并且降低下丘脑CRH mRNA与TNF-αmRNA的表达,对心衰大鼠产生干预影响。
OBJECTIVE: To observe the effects of Qili Qiangxin Capsule on the cardiac function, myocardial tissue morphology, neuroendocrine factor and hypothalamus in rats with chronic heart failure by microinjection pump. Methods: The model of heart failure was established by ligating the left anterior descending coronary artery in rats. After 4 weeks of modeling, Qiliqiangxin capsule solution (0.01mg / kg, 0.02mg / kg) Group). The positive control group was given CRH competitive inhibitor (αh-CRH). After 4 weeks of administration, carotid artery cannulation was used to detect the cardiac function indexes of rats in each group. RESULTS: 0.01mg / kg Qiliqiangxin Capsule solution and 0.02mg / kg Qizhenqiangxin capsule solution and CRH competitive inhibitor group could both improve the cardiac function and alleviate the changes of myocardial tissue morphology in rats with chronic heart failure, Reduce serum NT-pro BNP and plasma ACTH, PEG2, NE content, reduce hypothalamic CRH mRNA and TNF-αmRNA expression. CONCLUSION: QIQIANGXIN capsule administered by intracerebroventricular cannulation and microinjection pump can effectively improve cardiac function and myocardial tissue morphology in rats with heart failure, decrease neuroendocrine factor content, and decrease hypothalamic CRH mRNA and TNF-α mRNA The effect of intervention on heart failure rats.
