目的研究整合素αvβ3抑制剂RGDfv对骨肉瘤细胞增殖及凋亡的影响。方法通过MTT法检测不同浓度RGDfv序列肽对F5M2细胞生长增殖的抑制作用;通过Western blot方法观察经RGDfv诱导48 h的F5M2细胞中凋亡蛋白cleaved Caspase-3的表达。结果 MTT法检测结果显示经不同浓度RGDfv序列肽处理的F5M2细胞增殖受到抑制,OD值明显减少,并且呈浓度及时间依赖性(P<0.05 or 0.01);RGDfv处理后的F5M2细胞中,随着RGDfv浓度的升高,cleaved Caspase-3的表达量也较对照组明显增加(P<0.05)。结论 RGDfv序列肽能抑制F5M2细胞的增殖,并通过依赖于Caspase-3的通路诱导骨肉瘤F5M2细胞凋亡。
Objective To investigate the effects of integrin αvβ3 inhibitor RGDfv on proliferation and apoptosis of osteosarcoma cells. Methods MTT assay was used to detect the inhibitory effect of different concentrations of RGDfv peptide on the growth and proliferation of F5M2 cells. Western blot was used to observe the expression of cleaved Caspase-3 in F5M2 cells induced by RGDfv for 48 h. Results The results of MTT assay showed that the proliferation of F5M2 cells treated with RGDfv peptide of different concentrations was inhibited and the OD value was significantly decreased (P <0.05 or 0.01). In RGDfv-treated F5M2 cells, The increase of RGDfv concentration, cleaved Caspase-3 expression was also significantly increased compared with the control group (P <0.05). Conclusion RGDfv peptide can inhibit the proliferation of F5M2 cells and induce the apoptosis of osteosarcoma F5M2 cells through the Caspase-3-dependent pathway.
