目的:同时测定不同产地延胡索药材中3种主要生物碱(延胡索乙素、原阿片碱、去氢紫堇碱)成分的含量差异,并根据结果对药材质量进行科学分析。方法:采用Agilent HC-C_(18)(2)(250 mm×4.6 mm,5μm)色谱柱,以乙腈-0.1%磷酸水溶液(三乙胺调pH 5.3)(26∶74)为流动相,分析测定了全国各产地21批延胡索药材中3种生物碱成分的含量。使用SPSS 19.0软件对21批样品中3种生物碱成分含量进行聚类分析和主成分分析。结果:与全国延胡索主产地比较,安徽产延胡索药材中延胡索乙素含量均高于或接近道地及主流产区,聚类分析和主成分分析的分类结果也表明安徽产延胡索药材具有和主产地相同的品质。结论:安徽产延胡索药材质量上乘,具有良好的开发前景。聚类分析和主成分分析方法可为延胡索药材产地分类和质量评价提供依据,为合理开发利用药材资源提供参考依据。
OBJECTIVE: To simultaneously determine the contents of three main alkaloids (tetrahydropalmatine, protopine and dehydro-corydaline) in Corydalis yunnanensis rhizome from different areas and make scientific analysis on the quality of the medicinal materials. Methods: The mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (triethylamine adjusted to pH 5.3) (26:74) with Agilent HC-C18 (2) (250 mm × 4.6 mm, 5 μm) The contents of three alkaloids in 21 batches of Corydalis yanhusuo from various producing areas in China were determined. SPSS 19.0 software was used to cluster analysis and principal component analysis of three alkaloid components in 21 batches of samples. Results: Compared with the main producing areas of Corydalis yanhusuo, the contents of Corydamine in Corydalis yanhusuo in Anhui were higher than or close to those of the main producing areas. The classification results of cluster analysis and principal component analysis also showed that the Corydalis yanhusuo The same quality. Conclusion: The medicinal materials of Corydalis yannuo in Anhui have good quality and good development prospect. Clustering analysis and principal component analysis can provide the basis for the classification and quality evaluation of Corydalis yanhusuo medicinal materials, and provide a reference for the rational development and utilization of medicinal materials.
