用水囊引产2小时以内的5~7月龄胎儿大脑,按常规制备人胎脑胶质细胞(HFGC)作原代培养,并加入10mg/L LPS,经不同时相收获上清,作IL-6生物活性检测。结果表明,HFGC受LPS刺激后分泌IL-6的能力明显增加,5、6、7月龄胎儿IL-6的含量比自发分泌分别增加4.41倍、3.95倍和1.68倍;HFGC在换新鲜培养液后用LPS再刺激,仍可继续分泌高效价IL-6,培养5天时比非刺激组增加5.49倍,提示HFGC受LPS刺激后IL-6分泌能力增强。
Induction of water sac within 2 hours within 5 to 7 months old fetal brain, routine preparation of human fetal glial cells (HFGC) for primary culture and added 10mg / L LPS, harvested at different times the supernatant for IL- 6 biological activity test. The results showed that the ability of HFGC to secrete IL-6 after LPS stimulation was significantly increased, and the levels of IL-6 in embryos at 5, 6 and 7 months were 4.41 times, 3.95 times and 1.68 times higher than those in spontaneous secretion respectively. After stimulated with LPS, IL-6 could still continue to be secreted with high potency, which increased by 5.49-fold compared with non-stimulated group on day 5, indicating that IL-6 secreted by HFGC stimulated by LPS was enhanced.
