目的 培养人骨髓间充质干细胞 (mesenchymalstemcells,MSC)并检测其多能性。方法 抽取人胚胎股骨骨髓 ,Percoll分离液梯度分离后 ,将含MSC的低密度层培养于MSCGM培养基并扩增MSC。将MSC种植于裸鼠皮下 ,4周后观察MSC的分化情况。结果 成功地进行了人MSC的原代和传代培养。植入裸鼠体内后MSC可分化成骨、软骨、脂肪、骨骼肌、肌腱样组织等结构。结论 人MSC是一种具有多能性的干细胞
Objective To culture human mesenchymal stem cells (MSCs) and test their pluripotency. Methods Human embryonic femur bone marrow was extracted. After gradient separation with Percoll, the low density layer containing MSC was cultured in MSCGM medium and MSCs were expanded. MSC was implanted subcutaneously in nude mice and the differentiation of MSC was observed after 4 weeks. Results The primary and subculture of human MSCs was successfully performed. MSC implantation in nude mice can differentiate into bone, cartilage, fat, skeletal muscle, tendon-like structure. Conclusion Human MSC is a pluripotent stem cell
