目的建立高效毛细管电泳法(HPCE)同时测定不同批次板蓝根药材中表告依春、尿嘧啶、胸腺嘧啶、腺苷、次黄嘌呤、鸟苷、尿苷7种核苷类成分含量的方法。方法采用未涂渍标准熔融石英毛细管(75μm×64.5 cm,有效长度56 cm)为分离通道,60 mmol·L-1硼砂-15 mmol·L-1β环糊精(pH 9.6)为运行缓冲液,分离电压为20 kV,检测波长为254 nm,毛细管温度为25℃,压力进样为5 kPa×10 s。结果 7种核苷类成分的响应峰面积与其相应质量浓度的线性关系良好(r>0.999 4);加样回收率为97.15%~100.40%。实验表明,不同批次板蓝根药材中7种核苷类成分的含量有所差异。结论该方法简便、可靠,重复性较好,可用于板蓝根质量的评价和控制。
OBJECTIVE To establish a high performance capillary electrophoresis (HPCE) method for the simultaneous determination of 7 kinds of nucleosides in different batches of Radix isatidis according to the content of epirubicin, uracil, thymine, adenosine, hypoxanthine, guanosine and uridine. Methods Uncoated standard fused silica capillary (75 μm × 64.5 cm, effective length 56 cm) was used as the separation channel and 60 mmol·L-1 borax-15 mmol·L-1β cyclodextrin (pH 9.6) The separation voltage was 20 kV, the detection wavelength was 254 nm, the capillary temperature was 25 ℃, and the pressure injection was 5 kPa × 10 s. Results The linear relationship between the peak areas of seven nucleoside components and their corresponding mass concentrations was good (r> 0.999 4). The recoveries of the seven nucleosides were 97.15% -100.40%. Experiments show that different batches of Radix isatidis 7 kinds of nucleoside content of different ingredients. Conclusion The method is simple, reliable and reproducible. It can be used to evaluate and control the quality of Radix.
