目的 :探讨端粒酶亚单位表达水平与人胰腺癌发生的关系 ,以及其表达与端粒酶活性高低的相关性。方法 :应用RT PCR、DNA测序等技术研究端粒酶亚单位在胰腺癌中的表达情况 ,用ELISA法检测端粒酶活性。结果 :2 4例胰腺癌标本中hTERTmRNA表达阳性率为 83.3% ,而癌旁组织为 8.3% ,两者有显著性差异。hTERTmRNA的表达是人端粒酶活性表达的关键性因素。hTR及TP1mRNA在癌组织及癌旁组织中的表达无统计学差异 ,与端粒酶活性的表达无相关性。结论 :端粒酶参与了胰腺癌的发生 ;hTERTmRNA表达水平的上调可能在胰腺癌形成过程中起重要作用
Objective: To investigate the relationship between the expression level of telomerase subunit and the occurrence of human pancreatic cancer and the correlation between the expression of telomerase activity and telomerase activity. Methods: The expression of telomerase subunits in pancreatic cancer was studied by RT PCR and DNA sequencing. Telomerase activity was detected by ELISA. Results: The positive rate of hTERT mRNA expression in 24 cases of pancreatic cancer specimens was 83.3%, while that of paracancer tissues was 8.3%. There was a significant difference between the two. hTERTmRNA expression is the key factor of human telomerase activity. The expression of hTR and TP1 mRNA in cancer tissue and paracancerous tissues had no statistical difference, but had no correlation with the expression of telomerase activity. Conclusion: Telomerase is involved in the development of pancreatic cancer. Upregulation of hTERT mRNA expression may play an important role in the development of pancreatic cancer
