目的探讨中药补肾调冲方对卵巢早衰模型大鼠血清FSH、LH、AMH水平及TNF-α(肿瘤坏死因子α)、IFN-γ(干扰素γ)表达的影响,进而观察中药补肾调冲方对雷公藤多苷片致卵巢早衰的治疗作用。方法将60只SD雌性大鼠随机分为六组,每组10只。分别设为空白对照组、模型对照组、中药补肾调冲方低、中、高剂量组、西药对照组。除空白组外其余各组均用雷公藤多苷片造模14d,造模结束24h后各给药组给予补肾调冲方治疗36d。给药结束后,ELISA法检测大鼠血清AMH、FSH、LH水平,RTPCR法检测TNF-α,IFN-γmRNA的表达。结果中药补肾调冲方能显著提高血清AMH水平,降低血清FSH、LH的水平,提高卵巢对激素的敏感性,降调TNF-α,IFN-γmRNA的表达,抑制卵泡过早耗竭,与西药对比无明显差异。结论中药补肾调冲方可提高卵巢早衰大鼠卵巢储备能力,促进卵泡发育。提示中药补肾调冲方对卵巢早衰可能有治疗作用。
Objective To investigate the effects of Bushen Tiaochong Recipe (STGP) on serum levels of FSH, LH and AMH and the expression of TNF-α (tumor necrosis factor-α) and interferon-γ in premature ovarian failure model rats. Therapeutic effect of vinorelbine tablets on premature ovarian failure. Methods Sixty SD female rats were randomly divided into six groups of 10 rats. They were set as blank control group, model control group, traditional Chinese medicine Bushen Tiaochong Recipe low, middle and high dose group and western medicine control group respectively. In addition to the blank group, the rest of the groups were treated with Tripterygium glycosides tablet 14d, 24h after the completion of modeling, each treatment group was given Bushen Tiaochong Recipe for 36 days. Serum AMH, FSH and LH levels were detected by ELISA and TNF-α and IFN-γmRNA were detected by RTPCR. Results Bushen Tiaochong Recipe could significantly increase the level of serum AMH, lower the serum levels of FSH and LH, enhance the sensitivity of ovary to hormones, decrease the expression of TNF-α and IFN-γmRNA, and inhibit premature depletion of follicles, compared with Western medicine Significant differences. Conclusion Bushen Tiaochong Recipe can improve ovarian reserve in rats with premature ovarian failure and promote follicular development. Prompt Chinese kidney Bushen Tiaochong on premature ovarian failure may have a therapeutic effect.
