目的探讨Beclin-1在儿童急性淋巴细胞白血病细胞的表达及与临床分型等特征之间的关系。方法应用免疫组织化学SP法(streptavidin-perosidase,链霉素抗生物素-过氧化物酶连结法),检测Beclin-1在急性淋巴细胞白血病和正常人外周血白细胞的表达情况。结果 (1)Beclin-1在急性淋巴细胞白血病外周血和正常人外周血有核细胞的表达率分别为28.5%(6/21)和72.5%(29/40)。急性淋巴细胞白血病的表达明显低于正常人,两者有明显的差异(P<0.01)。(2)Beclin-1在男性患儿中的表达率为36.3%(4/11),在女性患儿的表达率为20%(2/10),两组之间的差异无统计学意义(P>0.05)。(3)临床分型L1中Beclin-1的表达率为33.3%(4/12),在L2型中为11.1%(2/9),两者之间比较差异无统计学意义(P>0.05)。结论 Beclin-1在急性淋巴细胞白血病外周血有核细胞中的表达低于正常人外周血有核细胞。
Objective To investigate the expression of Beclin-1 in children with acute lymphoblastic leukemia and its relationship with clinical features and so on. Methods The expression of Beclin-1 in peripheral blood leukocytes of acute lymphoblastic leukemia and normal human was detected by streptavidin-perosidase (streptavidin-peroxidase-linked method). Results (1) The expression rates of Beclin-1 in peripheral blood and normal peripheral blood of patients with acute lymphoblastic leukemia were 28.5% (6/21) and 72.5% (29/40), respectively. Acute lymphoblastic leukemia expression was significantly lower than normal, there was a significant difference between the two (P <0.01). (2) Beclin-1 expression was 36.3% (4/11) in males and 20% (2/10) in females, with no significant difference between the two groups ( P> 0.05). (3) The positive rate of Beclin-1 was 33.3% (4/12) in clinical classification L1 and 11.1% (2/9) in L2, there was no significant difference between them (P> 0.05 ). Conclusion The expression of Beclin-1 in peripheral blood mononuclear cells of acute lymphoblastic leukemia is lower than that of normal human peripheral blood mononuclear cells.
