目的 :观察牛磺酸 (Taurine)对甲状腺功能亢进 (以下简称甲亢 )豚鼠离体心脏窦房结细胞动作电位的变化 ,探讨牛磺酸对甲亢豚鼠离体心脏窦房结细胞电生理特性的影响及作用机制。 方法 :通过给豚鼠肌肉注射生理盐水 [或甲状腺素Na(L T4 Na) ]建立了对照组 (或甲亢组 )。 结果:(1)在 5、10、2 0mmol/L牛磺酸作用下 ,甲亢组豚鼠Vmax明显降低 ,SDR明显降低 ,APD50 、APD90 明显延长 ;(2 )在 10、2 0mmol/L牛磺酸作用下 ,对照组豚鼠Vmax、SDR明显降低 ,APD50 、APD90 明显延长 ;(3)Vmax、SDR在无牛磺酸作用下甲亢组比对照组明显增高 ,APD50 、APD90 明显缩短。 结论 :(1)牛磺酸抑制对照组豚鼠离体心脏窦房结细胞动作电位的Vmax、SDR ,可能与其具有钙拮抗作用有关 ;(2 )牛磺酸抑制甲亢组豚鼠离体心脏窦房结细胞动作电位的Vmax、SDR ,明显延长APD50 、APD90 ,可能与其调节心肌细胞内Ca2 + 稳态及促进K+ 外流有关。
Objective: To observe the change of action potentials of taurine in isolated sinus heart cells of guinea pigs with hyperthyroidism and to investigate the effect of taurine on the electrophysiological properties of sinoatrial node cells in guinea pigs with hyperthyroidism And mechanism of action. Methods: A control group (or hyperthyroidism group) was established by intramuscular injection of normal saline [or T4Na] to guinea pigs. Results: (1) The Vmax of guinea pigs in hyperthyroidism group decreased obviously and the SDR decreased significantly with the increase of taurine concentration of 5, 10, and 20mmol / L, while APD50 and APD90 prolonged obviously. (2) Vmax and SDR of guinea pigs in control group were significantly decreased and APD50 and APD90 were significantly prolonged; (3) Vmax and SDR were significantly increased in hyperthyroidism group without taurine compared with control group, while APD50 and APD90 were significantly shortened. Conclusion: (1) The Vmax and SDR of taurine in inhibiting the action potential of sinoatrial cells in isolated guinea pig hearts may be related to its calcium antagonism effect. (2) Taurine can inhibit the sinoatrial node Vmax and SDR of action potential of cells significantly prolonged APD50 and APD90, which may be related to its regulation of intracellular Ca2 + homeostasis and promotion of K + efflux.
