目的 :研究瘤体内直接注射脂质体包裹的人 IFNα基因的抗肿瘤效果。 方法 :在鳞癌荷瘤裸鼠的瘤体内注射体外已制备好的用脂质体包裹的 IFNα DNA,观察肿瘤生长状况及小鼠存活情况。结果 :瘤体内注射脂质体包裹的人 IFNα基因后 ,荷瘤裸鼠肿瘤的生长明显受抑 ,其中部分荷瘤裸鼠的肿瘤消失 ;荷瘤裸鼠的存活期明显延长 ,部分荷瘤裸鼠可长期存活。病理结果显示 ,治疗后的肿瘤细胞形状不一 ,有多数细胞质崩解 ,破坏明显。结论 :瘤体内注射脂质体包裹的 IFNα基因后 ,可在瘤体内原位直接转染肿瘤细胞并破坏该肿瘤细胞 ,从而达到抑制肿瘤生长的效果。
Objective: To study the anti-tumor effect of direct injection of human liposome-encapsulated human IFNα gene in vivo. METHODS : The liposome-encapsulated IFNα DNA was prepared in vitro in the tumor-bearing nude mice bearing squamous cell carcinoma to observe tumor growth and survival. RESULTS: After intratumoral injection of liposome-encapsulated human IFN alpha gene, the growth of tumor-bearing nude mice was significantly inhibited. The tumors of some tumor-bearing nude mice disappeared; the survival of tumor-bearing nude mice was significantly prolonged, and some tumors were bare. Rats can live long-term. The pathological results showed that the shape of the tumor cells after treatment was not uniform, and most of the cytoplasms disintegrated and destroyed significantly. Conclusion: After intratumoral injection of the liposome-encapsulated IFNα gene, tumor cells can be directly transfected into the tumor cells and destroy the tumor cells in order to inhibit tumor growth.
