目的观察由腺病毒载体介导的前尿激酶(proUK)基因导入家兔颈动脉后的表达和对血小板在损伤动脉内膜沉积的影响。方法将proUK基因腺病毒载体局部注射入家兔右颈总动脉腔内,用野生型腺病毒(Ad)局部注射入左颈总动脉腔内作为自身对照。用免疫组织化学染色观察proUK基因在血管内膜的表达;家兔血小板用111In标记并回输体内后,用电刺激损伤转基因血管内膜,定量观察111In标记血小板在损伤血管内膜沉积量,血管切片HE染色观察血栓形成情况。结果转Ad/proUK血管内皮细胞有明显的proUK阳性免疫反应颗粒,而转Ad的血管内皮未见;单位干重血管标记血小板数于转Ad/proUK血管[(4.60±0.93)×107/g]明显低于转Ad者[(27.95±4.93)×107/g](P<0.01);HE染色可见转Ad/proUK血管腔内只有少量血栓存在,而转Ad血管有大块血栓,几乎堵塞整个血管。结论局部血管内导入Ad/proUK后,可明显抑制血小板在损伤血管内膜的沉积,从而抑制动脉血栓形成。
Objective To observe the effect of adenovirus vector-mediated pro urokinase (proUK) gene on the carotid artery in rabbits and its effect on platelet damage in arterial intima. Methods The adenovirus vector of proUK gene was injected locally into the right common carotid artery in rabbits. The wild type adenovirus (Ad) was injected locally into the left common carotid artery as its own control. Immunohistochemical staining was used to observe the expression of proUK gene in the intima of blood vessels. After the platelets were labeled with 111In and transfused into the body, the transgene intima was injured by electrical stimulation. The amount of 111In-labeled platelet in injured intima, Slice HE staining observed thrombosis. Results The proUK-positive immunoreactive particles in Ad / proUK-transfected VECs were not found, but the number of transvascular Ad-provascular endothelial cells did not reach the level of Ad / proUK [(4.60 ± 0.93) × 107 / g] was significantly lower than that in Ad-transfected mice [(27.95 ± 4.93) × 107 / g] (P <0.01) Ad blood vessels have a large thrombus, almost plug the entire blood vessel. Conclusion The local intravascular administration of Ad / proUK can significantly inhibit the deposition of platelets in injured vascular intima, thus inhibiting arterial thrombosis.
