为研究三氧化二砷和亚砷酸钠抑制人肝癌BEL7404细胞及宫颈癌HeLa细胞生长时细胞超微结构的变化,调整细胞密度均为1×105个/ml,分别加入0(对照)、0.25、2.0μmol/L三氧化二砷溶液或亚砷酸钠溶液染毒7 d。在电镜下观察细胞的超微结构。结果显示,与对照组比较,2.0μmol/L三氧化二砷处理的HeLa细胞细胞凋亡小体形成率较高,差异有统计学意义(P<0.01)。2.0μmol/L亚砷酸钠和三氧化二砷染毒BEL7404细胞及HeLa细胞出现体积缩小,细胞内染色质浓集于核膜内面呈团块状,或染色体断裂成碎块状,细胞核固缩边缘化等典型的凋亡细胞特征性改变。许多细胞浆内有空泡形成。0.25μmol/L三氧化二砷溶液或亚砷酸钠染毒对BEL7404细胞及HeLa细胞均未见典型凋亡结构。提示高浓度(2.0μmol/L)亚砷酸钠和三氧化二砷可诱导肿瘤细胞凋亡。
