[目的]探讨不同浓度健脾解毒中药对间接原位移植肝癌裸鼠模型的抗癌作用及最佳药效浓度。[方法]采用人肝细胞癌细胞株Bel-7402间接原位移植,造雄性肝癌裸鼠模型90只,造模后24 h随机分9组,即不同浓度健脾解毒复方中药A、B、C、D、E、F、G组,替加氟207化疗(FT)组,0.85%氯化钠(NS)组,8周后处死裸鼠,观察各组荷瘤裸鼠的生存时间、体重变化、肿瘤形成情况、肝脾指数等。另外10只雄性裸鼠不造模作为正常对照(DZ)组。[结果]造模成功率为76.14%,实验结束获得有效病理标本67例,成瘤以多发小结节为主;各中药组的生存时间和生存率均优于FT组(P<0.05),中药D、E组的生存时间>NS组(P<0.05);各中药组荷瘤裸鼠治疗后体重有增加(P<0.05),FT组治疗后体重明显下降(P<0.05);中药组的肝指数和脾指数均>FT组(P<0.05)。[结论]健脾解毒复方中药可以增加荷瘤裸鼠的体重,改善营养状态,延长带瘤生存时间。健脾解毒复方中药抗癌作用的有效范围为7.5~20.0 g/kg。
[Objective] To investigate the anticancer effect and the best concentration of different concentration Jianpijiedudui Indirect Indwelling HCC nude mice model. [Method] Indirectly transplanted with human hepatocellular carcinoma cell line Bel-7402 in situ, 90 male HCC nude mice models were established. After 24 h, 24 nude mice were randomly divided into 9 groups, namely, different concentrations of Chinese herbs A, B, C , D, E, F and G groups, Tegafur 207 chemotherapy group (FT) and 0.85% sodium chloride group. After 8 weeks, the nude mice were sacrificed and the survival time and body weight , Tumor formation, liver and spleen index. Another 10 male nude mice were not modeled as a normal control (DZ) group. [Results] The successful rate of model making was 76.14%. There were 67 effective pathological specimens at the end of the experiment. Multiple nodules were the main tumorigenicity. The survival time and survival rate of each TCM group were better than those of FT group (P <0.05) The survival time of Chinese medicine group D and group E was higher than that of NS group (P <0.05). The body weight of nude mice in each TCM group increased (P <0.05), and the body weight of FT group decreased significantly (P <0.05) The liver index and spleen index were> FT group (P <0.05). [Conclusion] Jianpi Jiedu Compound Chinese herbal medicine can increase the body weight of tumor-bearing nude mice, improve the nutritional status and prolong the survival time of the tumor. The effective range of anti-cancer effect of Chinese medicine for invigorating the spleen and detoxifying herbs is 7.5 ~ 20.0 g / kg.
