目的:比较水煎煮、醇提取以及湿法超微粉碎3种提取工艺对全蝎中尿嘧啶、次黄嘌呤、黄嘌呤、鸟苷、腺苷5种核苷类成分含量的影响。方法:全蝎经不同工艺提取,采用HPLC外标法测定提取物中5种核苷化合物含量。Agilent C18色谱柱(4.6 mm×250 mm,5μm),流动相0.07%冰乙酸-甲醇梯度洗脱,流速1.0 mL·min-1,检测波长254 nm,柱温35℃。结果:尿嘧啶、次黄嘌呤、黄嘌呤、鸟苷、腺苷5种核苷化合物均在一定质量浓度范围内线性关系良好,r均>0.999 5,样品中5种核苷化合物的加样回收率98.9%~103.9%,RSD均<3%。结论:该HPLC简便快速、灵敏度高、重复性好,可应用于同时测定尿嘧啶、次黄嘌呤、黄嘌呤、鸟苷、腺苷5种核苷化合物含量并比较不同全蝎提取物之间的差异。
OBJECTIVE: To compare the effects of three decoction methods, water decoction, alcohol extraction and wet ultrafine pulverization on the contents of five nucleosides such as uracil, hypoxanthine, xanthine, guanosine and adenosine in Scorpio. Methods: Scorpio was extracted by different techniques. The contents of five nucleoside compounds in extracts were determined by HPLC external standard method. An Agilent C18 column (4.6 mm × 250 mm, 5 μm) with a mobile phase of 0.07% glacial acetic acid and methanol was used. The flow rate was 1.0 mL · min-1. The detection wavelength was 254 nm and the column temperature was 35 ℃. Results: Uracil, hypoxanthine, xanthine, guanosine, adenosine five kinds of nucleoside compounds in a certain concentration range of a good linear relationship, r> 0.999 5, samples of five kinds of nucleoside compound sample recovery Rates of 98.9% to 103.9%, RSD <3%. Conclusion: The HPLC method is simple, rapid, sensitive and reproducible. It can be used for the simultaneous determination of five nucleoside compounds, uracil, hypoxanthine, xanthine, guanosine and adenosine and to compare the contents of five different scorpion extracts difference.
