目的:研究槲皮素在诱导白血病K562细胞凋亡过程中PPARγ蛋白的表达。方法:用槲皮素作用于K562细胞一定时间后,通过胎盘蓝拒染法观察其生长曲线,MTT法检测其对细胞的抑制率,Hoechst33258染色后荧光显微镜下观察细胞核形态,流式细胞仪检测处理后细胞的凋亡率,采用Western-blotting技术从蛋白分子水平上检测槲皮素对PPARγ蛋白表达。结果:槲皮素显著抑制K562细胞的增殖并呈浓度依赖关系。电泳的结果显示一定浓度的槲皮素能够上调PPARγ蛋白的表达,进而可能诱导K562细胞的凋亡。结论:PPARγ蛋白表达的上调可能是槲皮素抑制K562细胞增殖诱导凋亡的原因之一,其中,槲皮素起着PPARγ非特异配体的作用。
Objective: To study the expression of PPARγ protein in the apoptosis of leukemia K562 cells induced by quercetin. METHODS: After treated with quercetin for K562 cells for a certain period of time, the growth curve was observed by placenta blue exclusion method. The inhibition rate of cells was measured by MTT assay. The nuclear morphology was observed under fluorescence microscope after Hoechst 33258 staining. Flow cytometry After treatment, the apoptosis rate of the cells was detected by Western-blotting technique from the molecular level of quercetin to PPARγ protein expression. Results: Quercetin significantly inhibited the proliferation of K562 cells in a concentration-dependent manner. The result of electrophoresis showed that certain concentrations of quercetin could upregulate the expression of PPARγ protein, which may induce the apoptosis of K562 cells. Conclusion: The up-regulation of PPARγ protein expression may be one of the reasons that quercetin inhibits proliferation-induced apoptosis in K562 cells. Among them, quercetin acts as a non-specific ligand for PPARγ.
