研究了AB-8大孔树脂对豆腐柴根提取物中总黄酮静态和动态的吸附性能。利用分光光度法测定样品中黄酮含量,考察提取液浓度、pH值、乙醇洗脱体积分数、上样液流速等实验条件对吸附结果的影响。结果表明,AB-8大孔树脂对豆腐柴根中总黄酮具有较好的吸附性能。静态吸附最佳条件:上样液浓度为0.0275mg/mL、pH 7、70%乙醇洗脱,总回收率为65.45%;动态吸附的最佳条件:浓度为0.0275mg/mL、pH为5、流速为1mL/min,90%的乙醇洗脱条件下,总回收率为47.27%。
The static and dynamic adsorption properties of AB-8 macroporous resin on the total flavonoids in the extract from Tofu was studied. The content of flavonoids in the sample was determined by spectrophotometry. The influences of the experimental conditions such as the concentration of the extract, the pH value, the elution volume fraction of ethanol and the flow rate of the sample on the adsorption were investigated. The results showed that AB-8 macroporous resin has good adsorption properties on the total flavonoids from the roots of tofu. The optimum conditions of static adsorption: the sample concentration was 0.0275mg / mL, pH7,70% ethanol, the total recovery was 65.45%. The optimum conditions for dynamic adsorption were as follows: the concentration was 0.0275mg / mL, pH was 5, The total recovery was 47.27% at a flow rate of 1 mL / min and 90% ethanol elution.
