目的:通过大黄虫丸对荷Lewis肺癌C57BL小鼠瘤体大小、生存期及环氧化酶2(cyclooxygenase,COX-2)表达影响的观察,初步探讨其抗肿瘤的可能机制。方法:造模后次日先随机分成5组,每组10只。据预试验结果确定高(H)、中(M)、低(L)剂量分别为0.18g/mL、0.09g/mL、0.045g/mL,以每只0.2mL灌胃10天;空白组小鼠以NS0.2mL灌胃10天;化疗组以50mg/kg CTX于第1天予腹腔给药。第11天处死小鼠,剖取完整肿块称重并计算抑瘤率。肿瘤组织以免疫组化、RT-PCR及western blot法检测COX-2的表达量。结果:①实验组高(H)、中(M)、低(L)和化疗组抑瘤率分别为29.70%、38.79%、17.58%、63.64%。实验高(H)、中(M)组与空白组比有统计学意义(P<0.05),低(L)与空白组比无统计学意义(P>0.05)。②实验组(M)、化疗组生存期分别为(19.14±0.77)、(19.80±0.75)天,显著长于空白组的(14.50±0.76)天(P<0.05);生命延长率分别为32.00%、36.56%,但实验组(M)与化疗组在生命延长作用方面无显著差异(P>0.05)。③免疫组化、RT-PCR及western-blot检测显示:大黄虫丸具有下调荷Lewis肺癌小鼠COX-2蛋白表达的作用,以实验组(M)作用最明显。结论:大黄虫丸对荷Lewis肺癌小鼠肿瘤具有明显的抑制作用并延长荷瘤小鼠生存期,作用机制可能与下调COX-2蛋白表达有关。
OBJECTIVE: To investigate the effect of Rhubarb Zhechong Pills on the tumor size, survival and the expression of cyclooxygenase 2 (COX-2) in Lewis lung carcinoma C57BL mice. Methods: The next day after modeling were randomly divided into 5 groups of 10. According to the results of pre-test, the high, middle and low doses of L were 0.18g / mL, 0.09g / mL and 0.045g / mL, respectively, The rats were gavaged with NS0.2mL for 10 days. The chemotherapy group was intraperitoneally administered with CTX 50mg / kg on the first day. Mice were sacrificed on the 11th day, the complete tumor mass was weighed and the tumor inhibition rate was calculated. Tumor tissues were detected by immunohistochemistry, RT-PCR and western blot COX-2 expression. Results: ① The inhibition rate of high, middle, low and chemotherapy in the experimental group were 29.70%, 38.79%, 17.58% and 63.64% respectively. There was a significant difference between experimental group (H), middle (M) group and blank group (P <0.05), and no significant difference between low (L) group and blank group (P> 0.05). ② The survival rates of experimental group (M) and chemotherapy group were (19.14 ± 0.77) and (19.80 ± 0.75) days respectively, significantly longer than those of the blank group (14.50 ± 0.76) days (P <0.05) , 36.56% respectively. However, there was no significant difference between the experimental group (M) and the chemotherapy group in life prolongation (P> 0.05). ③ Immunohistochemistry, RT-PCR and western-blot showed that Dahuang Shechong pill can down-regulate the COX-2 protein expression in Lewis lung cancer mice. The effect of experimental group (M) is the most obvious. Conclusion: The rhubarb capsule can significantly inhibit tumor growth in Lewis lung carcinoma mice and prolong the survival of tumor-bearing mice. The mechanism may be related to the down-regulation of COX-2 protein expression.
