目的探究大黄Rheum officinale提取物(ROE)对果蝇肠道干细胞(ISC)活性的影响。方法对照组果蝇食用正常玉米粉-酵母培养基,实验组果蝇食用含有0.05 g/mL或0.1 g/mL ROE的玉米粉-酵母培养基。对果蝇喂食有毒化合物诱导肠道损伤。分别检测分析果蝇的生存率、肠道前体细胞形态数目、ISC的增殖与分化、活性氧自由基(ROS)水平、肠上皮死亡细胞数目以及果蝇的寿命。结果 0.05 g/mL和0.1 g/mL ROE均可以提高肠道损伤后果蝇的生存率,0.1 g/mL ROE可以降低十二烷基硫酸钠(SDS)诱导后ROS的水平,减少死亡细胞的数目,抑制ISC的过度增殖与分化,缓解前体细胞的过度积累,维持肠道内稳态。此外ROE还可以延长果蝇寿命。结论 ROE可以抑制ISC的过度增殖和分化,增强果蝇肠道免疫功能,延长果蝇寿命。
Objective To investigate the effect of rhubarb Rheum officinale extract (ROE) on the intestinal tract stem cell (ISC) activity of Drosophila melanogaster. Methods The control flies consumed normal corn-yeast medium, while the experimental flies consumed corn-yeast medium containing 0.05 g / mL or 0.1 g / mL ROE. Feeding toxic compounds to fruit flies induces intestinal damage. The survival rate of Drosophila melanogaster, the number of intestinal precursor cells, the proliferation and differentiation of ISCs, reactive oxygen species (ROS) levels, the number of intestinal epithelial cells and the lifespan of Drosophila were detected. Results ROI at 0.05 g / mL and 0.1 g / mL could improve the survival rate of Drosophila after intestinal injury. 0.1 g / mL ROE could reduce the level of ROS induced by sodium dodecyl sulfate (SDS) and decrease the number of dead cells , Inhibit excessive proliferation and differentiation of ISC, relieve excessive accumulation of precursor cells and maintain intestinal homeostasis. In addition ROE can also extend the life span of fruit flies. Conclusion ROE can inhibit the excessive proliferation and differentiation of ISCs, enhance the intestinal immune function and prolong the life span of Drosophila melanogaster.
