目的探讨P63、Bcl-2、PCNA、Ki-67、MCMC2基因蛋白表达与成釉细胞瘤生物学行为的关系。方法60例成釉细胞瘤患者在河北医科大学口腔医院接受手术治疗。手术标本的存档蜡块及10例正常口腔黏膜标本,采用PV-9000免疫组化方法检测抗凋亡蛋白P63、Bcl-2,肿瘤增殖抗原PCNA、Ki-67、MCM2蛋白在成釉细胞瘤滤泡型和丛状型中的表达情况,进行定性及定量分析,并结合肿瘤包膜侵犯及临床复发情况进行探讨。结果 (1)P63的阳性表达在成釉细胞瘤滤泡型中低于丛状型(P<0.05),在正常组中的表达与滤泡型之间差异有显著性。P63在肿瘤的包膜是否侵犯病例及是否发生术后复发病例方面均具有显著性差异。(2)Bcl-2和PCNA在成釉细胞瘤滤泡型与丛状型中的表达分别与正常组之间差异有显著性。(3)成釉细胞瘤滤泡型中Ki-67的增殖指数和MCM2的阳性率均高于其在丛状型中的表达(P<0.05),且两型与正常组之间差异均有显著性。Ki-67和MCM2分别在术后复发病例与未复发病例之间比较差异有显著性。结论 (1)成釉细胞瘤P63的低表达与肿瘤包膜侵犯及复发密切相关。(2)成釉细胞瘤Ki-67和MCM2的高表达与肿瘤的复发密切相关。
Objective To investigate the relationship between the expression of P63, Bcl-2, PCNA, Ki-67 and MCMC2 protein and the biological behavior of ameloblastoma. Methods Sixty patients with ameloblastoma underwent surgery in Hebei Medical University Stomatological Hospital. Archival paraffin surgical specimens and 10 normal oral mucosa specimens, using PV-9000 immunohistochemical detection of anti-apoptotic proteins P63, Bcl-2, tumor growth antigen PCNA, Ki-67, MCM2 protein into the filter ameloblastoma Bubble and plexus in the expression, qualitative and quantitative analysis, combined with the tumor capsule invasion and clinical relapse situation to explore. Results (1) The positive expression of P63 in the ameloblastoma follicular type was lower than that in the plexus (P <0.05), and the difference between the expression of P63 and the follicular type was significant. P63 in the tumor capsule invasion of cases and whether the occurrence of postoperative recurrence cases have significant differences. (2) The expression of Bcl-2 and PCNA in follicular type and plexiform type of ameloblastoma was significantly different from the normal group. (3) follicular ameloblastoma in Ki-67 proliferative index of MCM2 and were significantly higher than the expression in the plexiform (P <0.05), and the differences between the two types are the normal group Significance. There was significant difference between Ki-67 and MCM2 in cases of recurrent and non-recurrent cases respectively. Conclusions (1) The low expression of P63 in ameloblastoma is closely related to tumor envelope invasion and recurrence. (2) The high expression of Ki-67 and MCM2 in ameloblastoma is closely related to tumor recurrence.
