为探讨 cyclin D1,cyclin E在乳腺癌发生发展中的作用及其与细胞周期调控相关基因蛋白的关系 ,采用免疫组化检测 17例非增生乳腺导管上皮、 19例不同程度增生的导管上皮及 5 9例乳腺癌中 cyclin D1,cyclin E,p16 ,p2 1waf1及 Rb基因蛋白的表达。结果显示 :1.非增生乳腺导管上皮除 1例 cyclin E过表达外均无 cyclin D1和 cyclin E的过表达。乳腺癌的 cyclin D1和 cyclin E的过表达率均明显高于良性乳腺组织 (P<0 .0 5 )。 2 .乳腺癌 cyclin D1过表达与淋巴结转移呈正相关 (P<0 .0 5 ) ,瘤体直径大于 5 cm者 cyclin E过表达呈增加趋势 ,但差异无显著性意义。 3.Cyclin D1和 cyclin E的过表达呈正相关 (P<0 .0 5 )。 4.从非增生乳腺导管上皮到增生直至乳腺癌 ,p16 ,p2 1与 cyclin D1,cyclin E含量的比值逐渐递减 ,而 p2 1含量高于 cyclin D1的乳腺癌体积小、淋巴结转移率低 (P<0 .0 5 )。 p2 1阳性率与 cyclin D1过表达呈正相关 (P<0 .0 1) ,也随 cyclin E的过表达呈上升趋势。 Rb基因蛋白的强表达与 cyclin D1过表达呈正相关 (P<0 .0 1)。结果表明 :CyclinD1和 cyclin E蛋白过表达频发于乳腺癌早期 ,它们可能与 p16、 p2 1waf1、 p Rb共同参与了乳腺癌的发生发展
To investigate the role of cyclin D1, cyclin E in the development of breast cancer and its relationship with cell cycle related gene proteins, immunohistochemistry was used to detect 17 cases of non-proliferative breast ductal epithelium, 19 cases of ductal epithelium with varying degrees of proliferation, and 5 The expression of cyclin D1, cyclin E, p16, p2 1waf1 and Rb protein in 9 cases of breast cancer. The results showed that: 1. None of the non-proliferative breast ductal epithelium overexpressed cyclin D1 and cyclin E except one case of cyclin E overexpression. The overexpression rates of cyclin D1 and cyclin E in breast cancer were significantly higher than those in benign breast tissue (P<0.05). 2. There was a positive correlation between cyclin D1 overexpression and lymph node metastasis (P<0.05). The overexpression of cyclin E in tumors with a diameter greater than 5 cm showed an increasing trend, but there was no significant difference. 3. The over-expression of Cyclin D1 and cyclin E was positively correlated (P<0.05). 4. The ratio of p16, p21 and cyclin D1 and cyclin E gradually decreased from non-proliferating breast duct epithelium to hyperplasia to breast cancer. The p21 content was higher than that of cyclin D1 and the lymph node metastasis rate was lower. <0 .0 5 ). The positive rate of p21 was positively correlated with the overexpression of cyclin D1 (P < 0.01), and it was also increased with the overexpression of cyclin E. The strong expression of Rb gene protein was positively correlated with the overexpression of cyclin D1 (P<0.01). The results showed that the overexpression of CyclinD1 and cyclin E proteins frequently occurred in early stage of breast cancer. They may be involved in the development of breast cancer with p16, p2 1waf1, and pRb.
