目的:探讨肿瘤坏死因子-α(TNF-α)和(或)白介素-1β(IL-1β)作用后小鼠腹主动脉的炎症反应及内皮P-选择素的表达。方法:32只小鼠随机平均分为3个实验组(IL-1β组、TNF-α组、TNF-α+IL-1β组)和1个对照组,实验组在小鼠腹主动脉周围无菌注射炎性因子TNF-α和(或)IL-1β(对照组用PBS),4h后取出腹主动脉行HE染色和内皮P-选择素免疫组化染色,定量分析动脉病理形态学的改变和半定量分析内皮P-选择素表达强度。结果:对照组动脉未见明显的病理改变及内皮P-选择素表达,3个实验组的动脉组织则发生不同程度的急性炎症改变,并以TNF-α+IL-1β组为甚,该组动脉发生的病理改变程度和内皮P-选择素表达的强度与范围明显大于IL-1β组和TNF-α组(P<0.05)。结论:联合应用TNF-α和IL-1β诱导的大动脉内皮的炎性病变稳定可靠,可用于急性冠状动脉综合症和相关大动脉内皮炎性病变早期的研究。
OBJECTIVE: To investigate the inflammatory reaction and the expression of endothelial P-selectin in the abdominal aorta of mice after the action of tumor necrosis factor-α (TNF-α) and / or interleukin-1β (IL-1β). Methods: Thirty-two mice were randomly divided into three experimental groups (IL-1β group, TNF-α group, TNF-α + IL-1β group) and one control group. RESULTS: There were no obvious pathological changes and P-selectin expression in the arteries of the control group. The arterial tissues of the three experimental groups showed different degrees of acute inflammatory changes, and were significantly different in the groups of TNF-α and IL-1β The extent of pathological changes of artery and the intensity and range of endothelial P-selectin expression were significantly higher than that of IL-1β and TNF-α groups (P <0.05). Conclusion: The combined application of TNF-α and IL-1β-induced inflammatory lesions in the aortic endothelium is reliable and can be used in the early stage of acute coronary syndrome and related aortic endothelium-induced inflammatory diseases.
