目的:研究参苓术汤(人参、白术、茯苓)对脾虚模型大鼠,环丙沙星药动学的影响。方法:24只大鼠随机分为4组,生理盐水组(给予生理盐水)、生理盐水+参苓术汤组(给予生理盐水和参苓术汤)、利血平组(给予利血平预处理)、利血平+参苓术汤组(给予利血平预处理后用参苓术汤治疗),以20mg/kg灌服环丙沙星,于给药后不同时间点采集血样,用高效液相色谱法分析环丙沙星的血浓度,根据血浓度-时间数据计算药动学参数。结果:利血平引起的大鼠症状类似于中医药临床脾虚证。环丙沙星在理盐水组、生理盐水+参苓术汤组、利血平组、利血平+参苓术汤组大鼠体内的药动学参数分别为:AUC_((0-∞))=(8.55±0.99),(7.41±1.39),(4.60±0.95)和(7.89±1.41) mg/(L·h),C_(max)=(3.31±0.63),(2.38±1.15),(1.29±0.45)和(3.35±1.15)mg/L。与生理盐水组相比,利血平组大鼠体内,环丙沙星的AUC_((0-∞))和C_(max)显著降低(P<0.01),与利血平组相比,利血平+参苓术汤组大鼠体内环丙沙星AUC_((0-∞))和C_(max)显著降低(P<0.01),而生理盐水组与生理盐水+参苓术汤组参数之间没有显著性差异。结论:脾虚状态下环丙沙星的生物利用度显著降低,参苓术汤可改善脾虚状态下环丙沙星的吸收。
Objective: To study the effect of Shenqishutang (Ginseng, Atractylodes macrocephala Koidz, Radix Astragali) on pharmacokinetics of ciprofloxacin in spleen deficiency model rats. METHODS: Twenty-four rats were randomly divided into 4 groups: normal saline group (administrated with normal saline), normal saline + Shenqizhu decoction group (administered with normal saline and Shenqi Shutong Decoction), and reserpine group (prepared with reserpine) Treatment), reserpine + Shenqi Shu Tang group (prepared by reserpine treatment after Shen Qi Shu Tang treatment), 20mg/kg ciprofloxacin fed, at different time points after the collection of blood samples, with The blood concentration of ciprofloxacin was analyzed by HPLC, and the pharmacokinetic parameters were calculated based on the blood concentration-time data. Results: The symptoms of reserpine-induced rats were similar to those of spleen deficiency syndrome in traditional Chinese medicine. The pharmacokinetic parameters of ciprofloxacin in the saline group, normal saline + Shenqishu decoction group, reserpine group, and reserpine + Shenqizhu decoction group were: AUC_((0-∞) )=(8.55±0.99), (7.41±1.39), (4.60±0.95) and (7.89±1.41) mg/(L·h), C_(max)=(3.31±0.63), (2.38±1.15), (1.29±0.45) and (3.35±1.15) mg/L. Compared with the saline group, the AUC_((0-∞)) and C_(max) of ciprofloxacin in the reserpine group rats were significantly decreased (P<0.01), compared with the reserpine group, The serum ciprofloxacin AUC_ ((0-∞)) and C_ (max) were significantly lower in Xueping + Shenqishu decoction group (P<0.01), while the normal saline group and normal saline + Shenqi Decoction decoction group parameters There was no significant difference between them. Conclusion: The bioavailability of ciprofloxacin is significantly reduced in spleen deficiency condition. Shenqishutang can improve the absorption of ciprofloxacin under the condition of spleen deficiency.
