目的:初步探讨凋亡“核心蛋白酶”———Caspase-3在槟榔碱(Arecoline)诱导血管内皮细胞(EndothelialCells,EC)凋亡中的作用。方法:体外培养人脐静脉内皮细胞(Hum an Umb ilicalVe in Endothelial Cells,HUVECs),将EC与槟榔碱共同培养,应用荧光显微镜观察EC凋亡情况,计算凋亡百分率并用比色法检测Caspase-3的活性。结果:槟榔碱诱导血管内皮细胞组在荧光显微镜下可见到明显的细胞凋亡改变,Caspase-3活性较正常对照组明显增高(P<0.05),且在12 h时最高。结论:槟榔碱可诱导血管内皮细胞凋亡,Caspase-3可能参与了这一细胞凋亡过程的调控。
AIM: To investigate the role of Caspase-3 in the apoptosis of endocervical cells (ECs) induced by arecoline. Methods: Human umbilical vein endothelial cells (HUVECs) were cultured in vitro. The ECs were co-cultured with arecoline. The apoptosis of ECs was observed by fluorescence microscopy. The percentage of apoptosis was calculated and the expression of Caspase-3 Activity. Results: The apoptosis of vascular endothelial cells induced by arecoline was observed under fluorescence microscope. The activity of Caspase-3 was significantly higher than that of normal control group (P <0.05), and it was the highest at 12 h. CONCLUSION: Arecoline can induce apoptosis of vascular endothelial cells. Caspase-3 may be involved in the regulation of apoptosis in this cell.
