采用本实验室自行制备的反相高效液相色谱柱,以醋酸钠缓冲液和乙腈-水线性梯度洗脱,在室温下对常见的16种1-氟-2, 4-二硝基苯(FDNB)衍生化氨基酸进行了有效的分离,用紫外吸收检测器(360nm)和外标定量法对灵芝蛋白中各种氨基酸的含量进行了测定。结果表明:FDNB衍生化氨基酸在一周内是稳定的,回收率在96%~102%之间,相对标准偏差小于4%,灵芝中含有人体必需的赖氨酸、苯丙氨酸和组氨酸等组分。
