目的 :观察转人TNFα基因对肾细胞癌细胞 (RCC)致瘤性的影响 ,为进一步对RCC的基因治疗打下基础。 方法 :将人TNFα基因构建入逆转录病毒载体 ,经包装、鉴定后 ,感染RCC细胞株 786 0 ,ELISA法测转人TNFα基因 786 0细胞上清中TNFα的活性 ,同时 ,将转人TNFα基因的 786 0细胞接种裸鼠 ,观测转TNFα前后RCC细胞致瘤性的变化。结果 :转人TNFα786 0细胞上清TNFα的浓度平均为 (5 0 0 4± 6 2 4) pg/ml,接种裸鼠后无肿瘤长出。结论 :转人TNFα基因使建株RCC失去了致瘤性。
Objective: To observe the effect of human TNFα gene transfer on tumorigenicity of renal cell carcinoma (RCC) cells and lay a foundation for further gene therapy of RCC. METHODS: Human TNFα gene was constructed into a retroviral vector. After packaging and identification, RCC cell line 786 0 was infected. TNFα activity in the supernatant of human TNFα gene 7600 cells was measured by ELISA. At the same time, human TNFα gene was transferred. 786 0 cells were inoculated into nude mice, and the tumorigenicity of RCC cells before and after transfection of TNFα was observed. Results : The average concentration of TNFα in the supernatant of transformed TNFα786 0 cells was (5 0 4 4 6 4) pg/ml, and no tumor grew after inoculation in nude mice. Conclusion : The transgenic human TNFα gene has lost its tumorigenicity to RCC.
