目的观察通心络超微粉(TXLU)对血管内皮功能的作用及机制。方法将健康雄性Wistar大鼠随机分为正常组和模型组,模型组以蛋氨酸饮食复合“基础饮食+负重游泳”法建立络气虚滞型血管内皮功能障碍动物模型。取正常组和模型组大鼠血清体外培养人脐静脉内皮细胞(HUVEC)细胞株,实验分为4组:空白组、正常血清组、模型血清组、TXLU组,每组4个复孔。观察血清作用HUVEC6h后细胞骨架蛋白纤维状肌动蛋白(F-actin)的形态、分布及含量变化。结果模型组血清作用6h后细胞骨架蛋白F-actin发生明显的破坏,其荧光强度减弱,TXLU预处理HUVEC后可减少F-actin损伤的发生及伴随含量的增加(P<0.01)。结论 TXLU可以减弱细胞骨架蛋白F-actin的损伤,可能是其改善血管内皮功能,防治动脉粥样硬化的机制之一。
Objective To observe the effect and mechanism of Tongxinluo superfine powder (TXLU) on vascular endothelial function. Methods Healthy male Wistar rats were randomly divided into normal group and model group. The animal model of vascular endothelial dysfunction was established by methionine diet combined with basal diet + weight-bearing swimming method. Cultured human umbilical vein endothelial cells (HUVEC) cell lines from normal and model rats serum were divided into 4 groups: blank group, normal serum group, model serum group and TXLU group. The changes of the morphology, distribution and content of cytoskeletal F-actin in HUVEC after 6 hours were observed. Results The serum F-actin in the model group was significantly damaged after 6 hours of treatment, and its fluorescence intensity was weakened. The pretreatment of HUVEC with TXLU could reduce the incidence of F-actin injury and the concomitant increase of the content of F-actin (P <0.01). Conclusion TXLU can attenuate the cytoskeletal F-actin damage, which may be one of the mechanisms of improving vascular endothelial function and preventing and treating atherosclerosis.
