本研究的目的是观察CD10在儿童期B 急性淋巴细胞白血病 (B ALL)中的表达情况 ,并探讨CD10在白血病微小残留病检测中的应用价值。用流式细胞术分析 5 8例儿童期急性淋巴细胞白血病表型 ,观察CD10在B ALL病例骨髓细胞中的表达特点 ,并将CD10抗体与其它多种抗体构成四色组合用于B ALL的微小残留病检测 ,评价其所起到的作用。结果表明 ,6 5 .5 % (38/5 8)的B ALL具有CD10过强表达 (CD10 bright)的特点 ,而且CD10 bright与CD34的阳性表达率有关———CD10 bright多出现于CD34阳性率高的B ALL中。CD10 bright在微小残留病检测中 ,与其它异常表达的抗原形成的组合能够在双参数点图上很好地区分出白血病细胞群与正常细胞群。即使在其他抗原表达无异常时 ,也可因CD10的过强表达而使白血病细胞群与正常细胞群区分开。结论 :CD10 bright与B ALL中CD34的高比例表达相关 ,CD10 bright也是微小残留病检测的一个很好的指标 ,CD10的表达与其他用于微小残留病检测的抗原表达的应用有互补作用。
The purpose of this study was to observe the expression of CD10 in childhood B acute lymphoblastic leukemia (B ALL) and to explore the value of CD10 in the detection of minimal residual disease of leukemia. Fifty-eight cases of acute lymphoblastic leukemia in childhood were analyzed by flow cytometry. The expression of CD10 in bone marrow cells of B ALL patients was observed. Four-color CD10 antibody and other antibodies were used in the minimal B ALL Residual disease testing, evaluation of its role. The results showed that 65.5% (38/5) of B ALL had the characteristics of CD10 bright, and the expression of CD10 bright was correlated with the positive rate of CD34 --- CD10 bright appeared in CD34 positive rate High in B ALL. CD10 bright In minimal residual disease testing, the combination with other abnormally expressed antigens makes a good distinction between leukemic and normal cell populations on a two-parameter plot. Even in the absence of abnormal expression of other antigens, leukemia cell populations can be differentiated from normal cell populations due to overexpression of CD10. CONCLUSION: CD10 bright is associated with a high expression of CD34 in B ALL, and CD10 bright is also a good marker for detection of minimal residual disease. CD10 expression is complementary to other antigens for detecting minimal residual disease.
