目的:研究银杏叶提取物抗肝纤维化作用及对CCl4肝纤维化大鼠RAS的影响。方法:采用CCL4造模的同时,分别给以银杏叶提取物及易善复混悬液灌服,共6周,光镜下观察病理改变;检测血清ALT、AST、ALP水平;HA、LN和血浆AngⅡ的水平;检测大鼠肝组织中α-SMA蛋白和AT1RmRNA表达。结果:银杏叶提取物可促使大鼠肝组织的炎症和纤维化程度减轻;使肝组织中血浆AngⅡ水平降低(P<0.01);AT1RmRNA、α-SMA蛋白表达降低(P<0.05,P<0.01)。结论:银杏叶提取物具有一定改善肝功能、抗肝纤维化的的作用。其抗肝纤维化作用的机制之一可能与其降低肝纤维化大鼠血浆AngⅡ水平,抑制肝组织AT1R的基因表达有关。
Objective: To study the anti-hepatic fibrosis effect of Ginkgo biloba extract and its effect on RAS in CCl4 hepatic fibrosis rats. Methods: The model of CCL4 was established. At the same time, Ginkgo biloba extract and Yixinte complex suspension were orally administrated for 6 weeks respectively. The pathological changes were observed under light microscope. The levels of serum ALT, AST and ALP, The level of plasma AngⅡ was measured. The expression of α-SMA protein and AT1R mRNA in rat liver tissue were detected. Results: Ginkgo biloba extract could reduce the degree of inflammation and fibrosis in rat liver tissue, decrease the plasma AngⅡ level in liver tissue (P <0.01), decrease the expression of AT1R mRNA and α-SMA protein (P <0.05, P <0.01) ). Conclusion: Ginkgo biloba extract has a certain effect of improving liver function and anti-liver fibrosis. One of the mechanisms of its anti-hepatic fibrosis may be related to its role in reducing plasma AngⅡ level and inhibiting AT1R gene expression in hepatic fibrosis rats.
